The research and development of new anticancer drugs has a large investment and long cycle, with an average development time of more than 10 years and an investment of more than 1.4 billion US dollars. In recent years, the extension of new indications for existing drugs (namely, Drug repurposture) is gradually receiving attention. Compared with the research and development of new drugs from scratch, "new use of old drugs" greatly shortens the research and development cycle and reduces the investment, which can effectively alleviate the shortage of drugs for cancer patients. We review the application of a series of non-tumor drugs in the treatment of tumors, and classify them into single or combined drugs according to ten characteristics of tumors. At the same time, we also systematically describe the application of experimental or computer based technologies such as thermal stability analysis, multiomics + bioinformatics in the "new use of old drugs". Finally, the problems and prospects of the strategy of "new use of old drugs" in the treatment of cancer are discussed and projected, emphasizing that "new use of old drugs" must have great potential in the era of precision medicine.
